Functional narrative

Seed maturation protein PM28 is one of the Late Embryogenesis-abundant Group IV (LEA IV) proteins. Function and mechanisms are not currently known, but they may play a protective role for membranes and proteins sensitive to dehydration. LEA IV proteins are also an important component of cellular glasses.

Modification type: Native
PDB:
Structural/functional type: Function arises via a molten globule to order transition
Functional classes: Chaperones
Functional subclasses: Unknown
Detection methods:
  1. Circular dichroism (CD) spectroscopy, far-UV (283 K; pH: 7; Potassium phosphate (buffer) 10 mM; Recombinant protein soln 10 uM; SDS (4%); Temp range 283K to 363K; TFE (50%))

  2. Fourier transform infrared spectroscopy (FTIR), aka infrared spectroscopy) (Aqueous lyophilized protein in D2O; Dry protein/sugar/poly-L-lysine/phospholipid (films for air-drying, 8 uL droplet of 20 ug/uL in H2O); Fast air-drying (RT, 3% RH, 3h); Slow air-drying (RT, 65% RH 16h + 3% RH, 3h); Temp range 233K to 423K)
