Functional narrative

Replication protein A (RP-A) is a three-subunit single- stranded DNA-binding protein (composed of RPA70, RPA32, and RPA14 subunits) with important functions in DNA replication, DNA repair and DNA recombination. The 70 kDa subunit of RP-A (hRPA70) contains an intrinsically unstructured linker domain (IULD, hRPA70 residues 105-182) that separates a protein interaction and weak ssDNA binding domain (DBD F, hRPA70 residues 1-104) from two high affinity ssDNA binding domains (DBD A and B, hRPA70 residues 183-422). DBD A, B, and F are structured domains that form five stranded anti-parallel beta barrels. The IULD functions to enhance the weak ssDNA binding affinity of DBD F, facilitating the competitive inhibition of DBD F-protein interactions.
