Functional narrative

The protein myosin is important in muscle contraction. It is located in the myofibril filaments of muscle tissue. Myosin consists of a lever arm and a head portion that binds weakly to actin, which is in a relatively fixed position. The lever arm of the myosin undergoes a conformational change which makes the head portion move in a swinging manner, known as a power stroke. These strokes cause very small movements approximately 10nm in length, which in turn cause the muscle to contract. The speed and strength of these contractions are believed to be dependent upon a 7 amino acid insert in the 25/50 kDa junction of myosin. Myosin containing the insert is capable of faster and stronger contractions than isoforms without the insert.

Comments:
The sequence information for this region came from Rovner et. al. (1997). It is a deviation from the Yanagisawa et. al sequence. This region is part of a 7 AA insert that helps to differentiate between fast and slow smooth muscle. The insert increases the protein’s enzymatic capabilities, as well as its ability to bind and move actin filaments.
